Abstract Fluorescence in situ hybridization (FISH) isa powerful technique to directly localize clonedDNA probes onto chromosomes. This approach isessential to delimit cytogenetic analysis, especiallyin karyotypic evolution study. In this review, thevarious applications of this method were summarized:chromosomal localization of repetitive DNA sequence,construction of chromosome mapping with singlecopy DNA sequences, cross-species chromosomepainting, comparative genomic hybridization andgenomic in situ hybridization, allowing a rapid andcomprehensive analysis of the process of chromosomalrearrangement in vertebrates. The compartmentaldata putting together from several applications ofFISH strategies provide insight into the likelyconstitution of a vertebrate ancestral karyotype andlead us to understand some of the intricacies ofkaryotypic evolution.
