| Abstract | Date palm (Phoenix dactylifera L.) is a dioecious plant with imperfect flower. The sex of plants can only be identified at the time of first flowering that takes approximately 3-7 years. This research aimed to screen primers for DNA markers of sex determination atthe seedling stage by Polymerase chain reaction (PCR) technique. The thirty-nine primerpairs previously reported and newly designed were selected and validated with known sexof date palm (KL1 and Bahee varieties) through PCR method with reference primer. Resultsshowed that newly designed primer pair, DpDOAmale5F and DpDOAmale4R could 100%identify male plants of KL1 and Bahee varieties. DNA fragment was presented only inmale plant and had a length of approximately 450 bp. The comparison of DNA extractionbetween homogenization with the metal bead and CTAB methods showed no difference ofthe DNA content and purity. However, the homogenization with the metal bead method couldextract faster. |
