| Abstract | Selected nitrofuran derivatives, specifically nitrofurazone, nitrofurantoin and furazolidone were determined by high performance liquid chromatography (HPLC) utilising a post-column chemiluminescence (CL) detection system. The CL reaction was based on the oxidation reaction between luminol and hydrogen peroxide in an alkaline medium and the enhancement of this reaction by nitrofurans. Optimum conditions were found: reagent stream of 1.5x10-3M luminol reagent prepared in a mixed solution of 0.01M sodium hydroxide and 1.0M sodium chloride; oxidant/micellar solution of 7.5x10-3M hydrogen peroxide in 0.1% Triton X-100; a mobile phase of acetonitrile : 10mM acetate buffer, pH 6.0 (1:9 v/v); injection loop of 200 uL; and analysis at room temperature. Under the optimal experimental conditions, linear calibration graphs of nitrofurazone, nitrofurantoin and furazolidone were obtained over concentration ranges of 0.1-5.0, 0.25-8.0 and 0.5-20.0 mg L-1 respectively. Detection limits were found to be 0.1, 0.25 and 0.5 mg L-1. The proposed HPLC-CL procedures were successfully applied to the determination of nitrofuran derivatives in pharmaceutical formulations, bee pollen and animal feeds. |
