| Abstract | Plant defensin family is one of the cysteine-rich antimicrobial peptides, which play important roles in plant immunity for responding against the invasion of pathogens. They are small cationic peptides which are shown to inhibit growth of a broad range of microorganisms. One of the major disease problems on production of cassava (Manihot esculenta Crantz) is cassava anthracnose disease, causing up to 80% yield loss by fungus Collectotrichum gloeosporiodes f. sp. manihotis (CAD). This research aims to identify and characterize defensin-related genes in cassava genome and investigated expression pattern of defensin genes upon CAD pathogen infection and to study the effect of phytohormone pre-treatment prior to fungal infection. The results of bioinformatics analysis showed that the coding sequences of 5 defensin gene members were identified in cassava genome and phylogenetic tree was constructed to verify each gene member. Also, defense-related response motifs were predicted in their promoter regions. Differential expression profiles of MeDef and MeDef1 . 2 were observed in CAD infection and phytohormone pre-treatment of the highly susceptible (HN) and less susceptible/ more resistant (HB60) cassava cultivars. The strong up-regulation of MeDef was found in HB60, whereas late induction of this gene was observed in HN. MeDef1 . 2 was not expressed. Salicylic acid pre-treatment prior to CAD infection could enhance defensive in HB60. This finding suggested that defensin has a potential function in CAD defense response in cassava, and further study should be useful for cassava improvement program to fungal pathogen resistance. |
