| Abstract | Longan (Dimocarpus longan Lour.) is commercially important crop in the northern part of Thailand. 'Do' variety has been popular, mostly grown about 79 % of growing areas. This causes low genetic diversity of longan. However, breeding program of longan typically takes a long time. Therefor, molecular markers could be developed to select F1hybrid. This study would develop SCAR marker for selection of F1hybrid of longan 'Do 27 X Si-Chomphu'. Fifty RAPD primers were screened and two primers (N-04 and BPS 5) showed polymorphic bands from Si-Chomphu. These bands were purified and cloned into the vector. Recombinant clones were sequenced. The DNA sequences were used for desining primer, which were specific to Si-Chomphu. The PCR products of these 2 primer pairs were 200 bp bands were used as the SCAR markers. These SCAR markers could distingnish Si-Chomphu and F1hybrid from Do 27 x Si-Chomphu |
