| Abstract | Pepper chat fruit viroid (PCFVd) is a mechanical and seed transmissible viroid. This work was to study mechanical inoculation methods and symptoms of PCFVd on Rutgers tomato. Seven mechanical inoculation methods were used, 1) PCFVd crude sap inoculation on leaves, 2) pressing on a leaf with PCFVd contaminated glove, 3) leaving contaminated glove 24 hrs under greenhouse condition before inoculation, 4) cutting an apical healthy stem with a PCFVd contaminated razor blade then take the cut-stem to plant, 5) cutting off an apical healthy stem with a PCFVd contaminated razor blade, 6) dipping a razor blade into PCFVd crude sap and doing a single slash in healthy plant stem, and 7) slashing the healthy plant stem 3-times with a PCFVd contaminated razor blade, uninoculated plants were used as control. The infected Rutgers tomato expressed apical shoot and plant stunt, necrosis of leaf vein, petiole and stem, yellow leaf tissue next to vein necrosis, leaf malformation, distorted petiole, small apical leave and some leaf-tip necrosis. Reverse transcription-polymerase chain reaction (RT-PCR) technique was used to confirm PCFVd infection at 4 and 8 weeks post-inoculation. Results showed that the transmission percentage of the 1st to 7th methods were 100, 100, 90, 20, 20, 20 and 70%, respectively. From this work, the 1st and 2nd inoculation methods were the best ones. |
