| Abstract | Major depressive disorder (MDD) is a serious health condition associated with neuroanatomical and functional alterations in many brain areas. As the active compound of rice bran oil, γ-oryzanol has been reported as having antioxidant, anti-inflammation, and neuroprotective effects, which protect the neurotransmitter deficits. Moreover, curcumin has been reported in antidepressant therapeutic properties. Therefore, this study aimed to determine the effects of γ-oryzanol combined with curcumin on dexamethasone-induced depressive-like behaviours in rats and hippocampal histological changes. Male Sprague-Dawley rats were randomly divided into 5 groups; Group 1, rats received reverse osmosis water (R.O.) as control, group 2 rats underwent dexamethasone-induced depression, group 3 and 4 depression-treated with γ-oryzanol combined with curcumin Formulation-1 (GOCur Form1) and Formulation-2 (GOCur Form2) respectively and group5, depression-treated with fluoxetine. All groups were evaluated the depressive–like behaviors by the forced-swimming and sucrose preference tests. Dexamethasone injection in the rats resulted in significant reductions in body weight, sucrose consumption and active time in the forced swim test (FST) when compared with control. Administration of GOCur Form1and GOCur Form2 reversed the depressive behaviours by increasing body weight and sucrose consumption and decreasing the immobility period. Furthermore, GOCur Form2 can restore histological changes in hippocampal damage from dexamethasone. These results indicate the potential of γ-oryzanol combined with curcumin for preventing and treating the depressive symptoms. |
