Title Identification of Genes Encoding Annexin in Cassava Genome and Expression Profile of MeAnn1 and MeAnn2 in Response to Pathogen Infection

Abstract Cassava (Manihot esculenta Crantz. ) is an important economic crop used as a raw material in food and various non-food industries. Nowadays, cassava production was limited by various diseases. Among several diseases, cassava anthracnose disease is one major fungal disease caused by Colletotrichum gloeosporioides f.sp. manihotis (CAD). Interestingly, there is a group of calcium-dependent membrane binding protein "Annexin", which can stimulate plant immunity upon pathogen infections. This study focused on identification and expression profiles of candidate annexin coding genes in cassava upon CAD infection. The bioinformatics study indicated that there were 10 cassava genes encoding annexin in which their promoter regions contained 28 cis-regulatory motifs related to plant immune system such as jasmonic acid ( JA) - responsive gene, salicylic acid ( SA) - responsive gene, resistance gene and others. The phylogenetic tree was constructed to verify each member of annexins and the protein structure was predicted. Disease Severity Rating suggested different resistant ability to CAD infection among the 4 cassava cultivars. The two cassava cultivars, Hanatee (HN, susceptible to CAD) and Huaybong60 (HB60, less susceptible to CAD) were selected to study. The differential expression of candidate genes encoding annexin was observed at different time points after CAD infection and in response to phytohormone (SA and JA) pre-treatment. The result showed possible function of cassava annexins related to defense response in CAD-susceptible and -less susceptible cultivars. This investigation on the role of annexins in relation with disease resistance in cassava will be useful for breeding program to improve cassava to resist against pathogen infection in future.
